Introduction of SARS-CoV-2

SARS-CoV-2, a newly identified coronavirus, has been spreading in China and has now reached multiple in other countries. The genetic analyses showed that SARS-CoV-2 emerged from a virus related to SARS and the possible symptoms include fever, cough, and shortness of breath. Based on the previous research results, these symptoms of SARS-CoV-2 may appear in 2-14 days after exposure. In this situation, public health officials, scientists, researchers, media and the public should take this current SARS-CoV-2 outbreak seriously and monitor it. Beta-D-N4-hydroxycytidine (NHC) Against Viruses

Beta-D-N4-hydroxycytidine (NHC), with the CAS No. 3258-02-4, is a base-modified ribonucleoside analogue recognized as a potent and selective antiviral candidate. NHC presents efficient antiviral activity against a variety of viruses, which include but are not limited to Chikungunya Virus (CHIKV), Venezuelan equine encephalitis Virus (VEEV), and Hepatitis C Virus (HCV).

Till now, the antiviral mechanism of action of NHC is still not completely understood. Some studies have shown that its nucleotide may act as a weak alternative substrate for CTP in the virus RNA polymerization reaction. In this case, insert the MHC-Monophosphate into viral RNA may change the thermodynamics of regulatory secondary structures. In conclusion, NHC might be a new efficient antiviral agent for the treatment of RNA virus infections. However, based on the stability studies in human and monkey whole blood, it showed that NHC might not persist enough time in plasma at a physiological pH and temperature to deliver significant antiviral levels of NHC into HCV-infected liver cells. In addition, a novel approach to prevent the oxidation or reduction of NHC should be developed for advancing this nucleoside to the clinic.
