Introduction of CPE

Various pathological cell alterations triggered by eukaryotic viruses are referred to as the CPE. Due to viral infections, the structure of the host cell changes and sometimes results in cell lysis. Common CPE includes rounding, detachment (plaques), clumping, ballooning (giant cell), fusion (syncytium formation), and inclusion body formation. The formation of inclusion bodies or syncytia induced by CPE can help with diagnosing viral infections. Therefore, by CPE, viral proliferation could be estimated through observable changes in cultured cells under a light microscope. In summary, CPE is a feasible scheme for viral isolation and identification.

Not only in the drug antiviral effect investigation, CPE assay was reported to be applied but also in methodology research of viral growth, titration, and neutralization of SARS-CoV-2 presented that is capable of the quantification of the neutralizing antibody titer in serum samples.

Effector Cell Vero E6 cells
Detection Scheme
  • Prepare 96-well plates of the desired cell line and incubate overnight;
  • 8 half-log serial dilutions in duplicate in the test medium;
  • Infected with the test virus diluted in the test medium;
  • Incubate at 37°C, 5% CO2 until CPE is apparent;
  • Stain with neutral red dye for 2 hours;
  • Add 200 µL 50:50 Sorensen citrate buffer/ethanol for >30 min., agitate, and read on a spectrophotometer at 540 nm;
  • Calculate EC50 and CC50 by regression analysis.
