Although the sensitivity and specificity of traditional antigen-antibody reactions are good, enzyme-linked immunoassay plays an important role in the detection of various biomolecules, but proteins as probe molecules are susceptible to environmental factors such as pH and temperature. But as a probe molecule, protein is easily denatured by environmental factors such as pH and temperature, and the synthesis is expensive. The aptamer is composed of DNA or RNA (mainly DNA), which is smaller than the protein. After screening and enrichment by SELEX, it can have sensitivity comparable to antigen-antibody reaction, and it is easier to synthesize and has better stability.
